APOBEC(“载脂蛋白质B mRNA编辑催化多肽”)是一类进化保守的胞苷脱氨酶家族。在人体内,已知含有保守的DNA胞嘧啶脱氨酶结构域的基因共有11种,包括AID、APOBEC1、APOBEC2、APOBEC3基因家族APOBEC3A、APOBEC3B、APOBEC3C、APOBEC3DE、APOBEC3F、APOBEC3G、APOBEC3H(分别称为A3A、A3B、A3C、A3D、A3F、A3G和A3H)和APOBEC4。APOBEC利用其脱氨酶活性通过与RNA和/或DNA结合,催化mRNA或使DNA中的胞嘧啶核苷酸转变为尿嘧啶,或者胞嘧啶核苷酸转变为胸腺嘧啶核苷酸,进而完成各自不同的功能。目前研究发现,AID及APOBEC3(A3s)的7种脱氨酶在人类的天然免疫和适应性免疫防御过程中发挥重要的作用,且在口腔癌,肺癌(腺癌和鳞状细胞癌),结直肠癌和乳腺癌等的诊疗过程中具有重要的潜在应用价值。AID可以通过将胞嘧啶脱氨基成尿嘧啶,来启动SHM (体细胞超突变)和CSR (类别转换重组),进而在抗体多样性方面发挥作用。它的异常表达能够使B细胞淋巴瘤等恶性肿瘤的发病频率显著增加。而A3A、A3B通过胞嘧啶到尿嘧啶转换,以及自身表达量上调而在乳腺癌和肺癌诊疗中起作用。A3G通过APOBEC3G/miR 29/MMP2为了解结直肠癌肝转移和开发治疗晚期结肠癌的有效疗法开辟了新的途径。综上所述,本文将以AID,A3A,A3B,A3G为例子,对APOBEC在癌症诊断和治疗方面的应用进行综述,以期为进一步药物研究和临床应用等提供参考。  相似文献

Seven major plastid protein encoding genes were positioned on the soybean chloroplast DNA by heterologous hybridization. These include the genes for the alpha, beta and epsilon subunits of the CF1 component of ATP synthase (atpA, atpB and atpE respectively), for subunit III of the CF0 component of ATP synthase (atpH), for the cytochrome f (cytF), for the ‘32 Kd’ thylakoid protein (psbA), and for the large subunit of ribulose-1,5-bisphosphate carboxylase-oxygenase (rbcL), all of which map in the large single copy region. The atpB, atpE and rbcL genes are located in the region adjacent to one of the segments of the inverted repeat. The genetic organization of the soybean chloroplast DNA is compared to that of other plastid genomes.  相似文献

The influence of sex steroid and pregnancy on the tissue concentration, uterine motor effect and receptor binding of VIP has been studied in the female genital tract of pregnant rabbits and oophorectomized rabbits during progesterone and/or oestrogen substitution. The concentration of immunoreactive VIP was high in the vagina and cervix, and lower in the uterine body of both pregnant and non-pregnant rabbits. A significant decrease in the VIP concentration (pmol/g wet weight) of the uterine body was observed toward term of pregnancy. The total uterine content of VIP, however, seems unchanged. Treatment of oophorectomized rabbits with ovarian steroids had no effect on the VIP concentration. The sensitivity for and potency of VIP on the relaxation of uterine muscle was significantly higher in oophorectomized rabbits treated with a combination of progesterone and oestrogen than in control rabbits. No difference was observed between non-pregnant and pregnant rabbits. The degradation and binding affinity for 125I-labelled VIP was highest in oophorectomized rabbits substituted with both oestrogen and progesterone. In the pregnant rabbits, the amount of receptors was decreased near term. In conclusion, sex steroids are able to influence the motor effect of VIP at the receptor level, but have no effect on the VIP concentration in the female genital tract.  相似文献

The effect of human pancreatic polypeptide (HPP) on rat pancreatic acini has been studied. It was found that HPP stimulated amylase and lipase release from the acini. The secretory response of acini to HPP was dose-dependent in a sigmoidal fashion. Between 10(-9) M and 10(-8) M concentration of HPP there was a slow increase of enzyme release to about 40-60% over basal release. At concentrations of HPP above 10(-8) M there was a rapid increase of enzyme release, amounting to 4-6 times over basal release at 10(-6) M concentration of HPP. The potency of HPP compared to other secretagogues at 10(-7) M concentration was 45% of CCK, 60% of carbachol and 75% of secretin. HPP did not inhibit the effect of CCK, secretin and carbachol on amylase release. The amylase release stimulated by HPP was accompanied by an increase in 45Ca2+ efflux. Atropine or dibutyryl cyclic GMP did not influence the effect of HPP. It is concluded that HPP stimulates the release of enzymes from rat pancreatic acini and that Ca2+ may be a mediator for this secretion.  相似文献

Isolated thylakoid membranes of Chlamydomonas reinhardi Y-1 with the 32-kDa polypeptide either radioactively labelled or unlabelled were incubated in vitro under various conditions in order to gain information about the degradation of the 32-kDa polypeptide. The degradation was higher at pH 6 compared with pH 7 and pH 8 and exhibited a temperature maximum between 20° C and 25° C (pH 6, pH 8). A light-dependent part of the total degradation was linearly dependent on white light of energy fluence rate between 1 and 20 mW·cm-2 at 25° C and leveled out at higher fluence rates. The degradation in light was only slightly stimulated by ATP but was reduced by 3-(3-4-dichlorophenyl)-1,1-dimethylurea. Adenosine-5-diphosphate and heparin (2.7 mM and 200 g per 100 l, respectively) known to inhibit kinases, caused a 50% decrease in degradation indicating that a phosphorylation step is involved in degradating the 32-kDa polypeptide. Out of various inhibitors specific for different types of proteases, only those for thiol- and endoproteases showed intense effects. These results point to a proteolytic degradation of the 32-kDa polypetide by a thylakoid-membrane-bound thiol-endoprotease. Its activity yields soluble breakdown products with relative molecular masses (Mrs) of 23, 16.5, 11.3 and 10.7 kDa, and these are accumulated in the in-vitro system. Partial proteolytic digestion of thylakoids with Staphylococcus aureus V8 protease results in at least two labelled breakdown products (Mrs 23, and 16.5 kDa). It is assumed that cleaving at identical amino-acid residues of the 32-kDa polypeptide by the thylakoid-membrane-bound thiolendoprotease and the V8 protease results in these two breakdown products. The fecundity, reproductive rates, and adult survival of Aphidius sonchi Marshall (Hymenoptera: Aphidiidae) parasitizing second and third instar nymphs of the sowthistle aphid, Hyperomyzus lactucae (L.) (Homoptera: Aphididae) were measured at six different host densities under constant laboratory conditions. At host densities of less than 50 aphids per flowering shoot per female per day, oviposition constraints resulting from the lack of hosts reduced the number of eggs laid, enhanced the extent of superparasitization and, as a result, effectively lowered the fecundity and reproductive rates of the parasites. Above this host density the parasites laid on average 220–230 eggs, but the effective fecundity and reproductive rates continued to increase with the host density. By contrast, the survivorship of the parasites seemed unaffected by host density, with an average adult life span of 4–5 days at all densities. Analysis of the data showed that the intrinsic rate of increase (rm) of the parasite varied with the host density and could reach values higher than that of the host under identical conditions. The response of rm to changes in host density and parasite sex ratio is illustrated.Overall, A. sonchi showed a typical convex functional response, to host density. However, the response showed obvious changes through the parasite's adult life and, furthermore, the rates of changes were not consistent at all host densities. The frequency distributions of parasite eggs were generally indistinguishable from random, and the number of hosts parasitized were predicted satisfactorily by the random oviposition equation.
Résumé L'étude a porté sure l'influence de 6 densités différentes d'Hyperomyzus lactucae (L.) (Homoptera: Aphididae), en conditions constantes de laboratoire, sur la fécondité, le taux de reproduction et la survie des adultes d'Aphidius sonchi Marshall (Hym. Aphidiidae, parasite des larves de 2e et 3e stades. A des densités inférieures à 50 pucerons par tige fleurie de Sonchus oleraceus L, par femelle et par jour, la limitation de la ponte due à l'absence d'hôtes a réduit le nombre d'oeufs émis, élevé le taux de superparasitisme et, en conséquence, diminué la fécondité et le taux de reproduction des parasites. Aux densités d'hôtes supérieures, les parasites ont pondu, en moyenne, 220 à 230 oeufs, mais la fécondité réelle et les taux de reproduction ont continué à augmenter avec la densité des pucerons. Par contre, la longévité des parasites n'a pas été affectée par la densité des hôtes, avec une durée moyenne de vie de 4 à 6 jours. L'analyse des données a montré que le taux d'accroissement intrinsèque (rm) du parasite a changé avec la densité des hôtes, et pourrait atteindre des valeurs supérieures à celles de l'ôte sous des conditions identiques. Les réponses de rm aux changements de densité des hôtes et au taux sexuel du parasite sont expliquées.Globalement, A. sonchi a présenté une réponse fonctionnelle convexe typique à la densité des hôtes. Cependant, cette réponse a changé au cours de la vie des images et, de plus, les taux de changement ne sont pas logiques à toutes les densités d'hôtes La fréquence de distribution des oeufs n'est généralement pas séparable d'une distribution au hasard, et le nombre d'hôtes parasites peut être prédit d'une façon satisfaisante en utilisant une équation de ponte au hasard.

Some streams near Dorset in south-central Ontario suffer from acid precipitation via run-off and seepage from thin soils with little buffering capacity. A spring-summer survey of eight headwater streams revealed some characteristics of their insect communities which could be correlated with pH. The streams could be divided into three groups according to pH and community structure. In the most acid group (annual pH range 4.3–4.8), Ephemeroptera were absent from two streams although mature Leptophlebia were collected just after spring thaw from the most acid one (pH 4.3–4.5). One of these three streams also lacked Plecoptera but the others had two or three genera, all shredders. The second group of three streams (pH 5.0–6.3), with one exception, did support Ephemeroptera (3–4 genera) and Plecoptera (1–4 genera), most of the latter being shredders. In all six of these acid streams, Trichoptera were more diverse and more dense than Ephemeroptera and Plecoptera; again, shredders were clearly dominant, especially the limnephilid caddisfly, Frenesia difficilis (Walker). These six streams also had similar chironomid communities (densities were an order of magnitude higher than other insects). Dominance by Chironomini and abundant Tanypodinae typified the most acid streams. In contrast, the two streams in the third group (pH 5.3–6.7) had richer and more balanced communities in general with relatively fewer shredders (no Frenesia), more collectors, and fewer Chironomini and Tanypodinae. As a field experiment showed that autumn-shed leaves decomposed more slowly in acid than in non-acid streams, summer-growing shredders may benefit from the pulse of acidity at snowmelt.  相似文献

A possible link between cellular cyclic AMP content and Na+K+ATPase activity was investigated in homogenates of rat kidney. Enzyme kinetics of Mg2+ and Na+K+ATPase were run in the presence of cyclic AMP, dibutyryl cAMP and compounds expected to elevate cyclic AMP levels such as forskolin, a potent adenylate cyclase activator, IBMX, an inhibitor of phosphodiesterases, and the beta-agonist isoproterenol. Medullary Na+K+ATPase is strongly inhibited by cyclic AMP whereas cortical Na+K+ATPase was stimulated in the same conditions. The correlation between ATPase activity and cellular cyclic AMP content supports the concept of a possible regulation of the enzyme by cyclic AMP.  相似文献

Although it is common knowledge that many trilobites enrolled, behavioral and functional aspects of enrollment are not at all well understood. Taphonomic details portrayed by enrolled trilobites in the Middle Devonian Hamilton Group (New York State) indicate that enrollment was a complex and morphologically constrained behavior. The trilobites Phacops rana (Green) and Greenops boothi (Green) are frequently enrolled in Hamilton strata; biostratinomic data indicate two very different enrollment postures. Interlocking morphologies (coaptative devices) and apodeme structure and disposition indicate that these postures reflect specific behaviors which involved interaction between tergal structures, inferred musculature, and the substratum. Phacops enrolled by burrowing forward and down into the sediment; dorsal muscles, attached to prominent articulating half-rings, imbricated the thorax such that each lappet overlapped the next posterior segment and locked into a posterior pleural facet. The pygidium was brought into place as the posterior segments of the thorax were placed into vincular notches along the lateral margin of the ventral cephalon. The pygidium locked with the cephalic vincular furrow to complete ‘perfect sphaeroidal’ closure. Greenops enrolled with the cephalon in an upright position at the sediment surface; a submarginal furrow on the ventral surface of the pygidium received the anterior rim of the cephalon. Relatively narrow articulating half-rings limited pleural rotation. Segments were loosely locked into narrow facets at the anterior margin of the next posterior lappet. In spite of rudimentary lappet and half-ring structures, Greenops displays an elaborate system of thoracopygidial muscles which involved dorsoventral and longitudinal attachments along the thorax and into the pygidium. Phacops, in contrast, displays very poorly developed apodemes which occur in the thorax only. Longitudinal muscle strength was likely less important during Phacops enrollment than is evident for the Greenops enrollment procedure. Conversely, Phacops clearly relied to a great degree upon competent closure devices which are poorly developed in Greenops. Biostratinomic data reveal different enrollment behaviors which reflect the function of different enrollment-related morphologies present in each species.  相似文献
